The ssDNA library (500 pmol) was pre-incubated for 3 min at 90 °C and immediately cooled for 1 h at 4 °C in 100 μL of binding buffer (20 mM Tris–HCl, 50 mM NaCl, 5 mM KCl, 5 mM MgCl2, pH 8.0) to stabilize the naturally occurring tertiary structure.

Refer to the aptamer reference source for the appropriate refolding parameters and binding conditions. Note: it is unknown whether aptamer functions properly without refolding.
Similar structured small antibiotics amoxicillin and benzylpenicillin do not bind to the aptamer.
